Novel acetylcholinesterase inhibitors

For more than two decades, the development of potent acetylcholinesterase (AChE) inhibitors has been an ongoing task to treat dementia associated with Alzheimer’s disease and improve the pharmacokineticproperties of existing drugs. Recently, Benoit David, Pascal Schneider, Philipp Schäfer were able to identify 11 new acetylcholinesterase inhibitors with low IC50 values (as part of the joint project CKB), one of which has a 5-fold lower inhibitory concentration than the drug already in use, rivastigmine. To identify potential candidates, they used three docking-based screening methods and tested a total of 61 compounds.
