In a joint publication, Diana Amariei and Mona Haase have presented two assays for the high-throughput analysis of enzymatic reactions. The indole assay is based on the coupling of DMAB to the indole ring in the presence of alcohols or acids and detects the degradation of the substrate. The pyrroloindole assay, on the other hand, detects the formation of the product with the aid of the CAM stain used in thin-layer chromatography.
Both methods can be used for the detection of natural products as well as for future investigations of pyrroloindole-forming enzymes.
